ELISA的基本原理有三條： 
    （1）抗原或抗體能以物理性地吸附于固相載體表面，可能是蛋白和聚苯乙烯表面間的疏水性部分相互吸附，并保持其免疫學活性；
    （2）抗原或抗體可通過共價鍵與酶連接形成酶結(jié)合物，而此種酶結(jié)合物仍能保持其免疫學和酶學活性； 
    （3）酶結(jié)合物與相應抗原或抗體結(jié)合后，可根據(jù)加入底物的顏色反應來判定是否有免疫反應的存在，而且顏色反應的深淺是與標本中相應抗原或抗體的量成正比例的，因此，可以按底物顯色的程度顯示試驗結(jié)果。

ELISA法是免疫診斷中的一項新技術(shù)，現(xiàn)已成功地應用于多種病原微生物所引起的傳染病、寄生蟲病及非傳染病等方面的免疫診斷。也已應用于大分子抗原和小分子抗原的定量測定，根據(jù)已經(jīng)使用的結(jié)果，認為ELISA法具有靈敏、特異、簡單、快速、穩(wěn)定及易于自動化操作等特點。不僅適用于臨床標本的檢查，而且由于一天之內(nèi)可以檢查幾百甚*千份標本,因此，也適合于血清流行病學調(diào)查。本法不僅可以用來測定抗體，而且也可用于測定體液中的循環(huán)抗原，所以也是一種早期診斷的良好方法。因此ELISA法在生物醫(yī)學各領(lǐng)域的應用范圍日益擴大，可概括四個方面： 1、免疫酶染色各種細胞內(nèi)成份的定位。 2、研究抗酶抗體的合成。 3、顯現(xiàn)微量的免疫沉淀反應。 4、定量檢測體液中抗原或抗體成份。
