人突觸素（SYP)ELISA Kit代測

ELISA的基礎(chǔ)是抗原或抗體的固相化及抗原或抗體的酶標(biāo)記。結(jié)合在固相載體表面的抗原或抗體仍然保持其免疫學(xué)活性，酶標(biāo)記的抗原或抗體既保留其免疫學(xué)活性，又保留酶的活性。在測定時，受檢標(biāo)本（測定其中的抗體或抗原）與固相載體表面的抗原或抗體起反應(yīng)。用洗滌的方法使固相載體上形成的抗原抗體復(fù)合物與液體中的其他物質(zhì)分開。再加入酶標(biāo)記的抗原或抗體，也通過反應(yīng)而結(jié)合在固相載體上。此時固相上的酶量與標(biāo)本中受檢物質(zhì)的量呈一定的比例。加入酶反應(yīng)的底物后，底物被酶催化成為有色產(chǎn)物，產(chǎn)物的量與標(biāo)本中受檢物質(zhì)的量直接相關(guān)，所以可根據(jù)呈色的深淺進(jìn)行定性或定量分析。由于酶的催化效率很高，間接地放大了免疫反應(yīng)的結(jié)果，使測定方法達(dá)到很高的敏感度。

ELISA試劑盒所需要樣本處理及要求 組織標(biāo)本：切割標(biāo)本后，稱取重量。加入一定量的PBS，PH7.4。用液氮迅速冷凍保存?zhèn)溆?。?biāo)本融化后仍然保持2-8℃的溫度。加入一定量的PBS(PH7.4)，用手工或勻漿器將標(biāo)本勻漿充分。離心20分鐘左右(2000-3000轉(zhuǎn)/分)。仔細(xì)收集上清。分裝后一份待檢測，其余冷凍備用。
ELISA試劑盒所需要樣本處理及要求  標(biāo)本采集后盡早進(jìn)行提取，提取按相關(guān)文獻(xiàn)進(jìn)行，提取后應(yīng)盡快進(jìn)行實驗。若不能馬上進(jìn)行試驗，可將標(biāo)本放于-20℃保存，但應(yīng)避免反復(fù)凍融.
