Anti-PCDH8抗體,原鈣粘附蛋白8抗體產(chǎn)品介紹：Protocadherins are a large family of cadherin-like cell adhesion proteins that are involved in the establishment and maintenance of neuronal connections in the brain. There are three protocadherin gene clusters, designated alpha, beta and gamma, all of which contain multiple tandemly arranged genes. PCDH8 (protocadherin-8), also known as Arcadlin or PAPC, is a 1,070 amino acid single-pass type I membrane protein that contains six cadherin domains and belongs to the protocadherin family. Localized to the cell membrane and expressed specifically in fetal and adult brain, PCDH8 is thought to play a role in cell adhesion events in the central nervous system (CNS). PCDH8 is inactivated or silenced in breast cancer, suggesting a possible role in tumor suppression. Two isoforms of PCDH8 that differ in their cytoplasmic tails are expressed due to alternative splicing events.
抗原修復(fù)，必須注意以下問題：
1、修復(fù)液的選擇。
0.01M枸櫞酸（pH6.0）：應(yīng)用zui多的抗原修復(fù)液，適用于大多數(shù)的抗原，用該液修復(fù)后的抗原表達(dá)增強(qiáng)，抗原的定性和定位很理想，結(jié)果不錯;
0.05M EDTA（pH8.0）：應(yīng)用較多的抗原修復(fù)液，修復(fù)能力較強(qiáng)，對于保存時間較長的切片或目的蛋白表達(dá)較弱的組織有很好的修復(fù)效果，需要控制好修復(fù)時間，否則容易出現(xiàn)較重的背景;
0.01M TBS（pH7.4）：中性修復(fù)液，適用于大多數(shù)抗原，對于核定位蛋白有較好的修復(fù)效果;
*：應(yīng)用較多的抗原修復(fù)液，適用于大多數(shù)的抗原，使用溫度37℃，對于容易脫片的切片有很好的保護(hù)作用。

2.抗原熱修復(fù)時應(yīng)選擇的溫度。
據(jù)實(shí)驗(yàn)認(rèn)為溫度在92℃－98℃是合適的，尤其在95℃為，這是因?yàn)椋海?）這種溫度未達(dá)沸騰，切片不容易脫離載玻片；（2）能夠解離和破壞與蛋白交聯(lián)的甲醛醛鍵等，處理時可以隨意選擇和確定抗原修復(fù)的作用時間。如果選擇高壓修復(fù)，因?yàn)闇囟容^高，時間不宜過長
3.抗原修復(fù)時有效溫度所需持續(xù)時間根據(jù)各單位的設(shè)備條件以及使用的儀器不同，抗原修復(fù)時在有效的溫度范圍內(nèi)所持續(xù)的時間也不一樣。
4.抗原修復(fù)液必須遵循自然降溫規(guī)律，否則效果不好或達(dá)不到抗原修復(fù)的目的。
5.盡量使用足量的抗原修復(fù)液，防止切片干涸。
6.切片必須附貼牢固，否則發(fā)生掉片。
