保存條件: Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.
Anti-GPI抗體,糖磷脂酰肌醇抗體產品介紹:This gene belongs to the GPI family whose members encode multifunctional phosphoglucose isomerase proteins involved in energy pathways. The protein encoded by this gene is a dimeric enzyme that catalyzes the reversible isomerization of glucose-6-phosphate and fructose-6-phosphate. The protein functions in different capacities inside and outside the cell. In the cytoplasm, the gene product is involved in glycolysis and gluconeogenesis, while outside the cell it functions as a neurotrophic factor for spinal and sensory neurons. Defects in this gene are the cause of nonspherocytic hemolytic anemia and a severe enzyme deficiency can be associated with hydrops fetalis, immediate neonatal death and neurological impairment.
免疫組化操作流程 (一) 儀器設備 1. 18cm不銹鋼高壓鍋或電爐或用微波爐. 2. 水浴鍋 (二) 試 劑 1. PBS緩沖液(ph7.2―7.4):NaC137mmol/L,KCl 2.7mmol/L ,Na2HPO4 4.3mmol/L, KH2PO4 1.4mmol/L. 2.0.01mol/L檸檬酸鈉緩沖液(CB,ph6.0,1000ml):檸檬酸三鈉3g,檸檬酸0.4g。 3.0.5mol/L EDTA緩沖液(ph8.0):700ml水中溶解186.1g EDTA& 8226;2H2O,用10mmol/L NaOH調至ph8.0,加水至1000ml. 4. 1mol/L的TBS緩沖液(ph8.0):在800ml水中溶解121gTris堿,用1N的HCl調至ph8.0,加水1000ml。 5. 酶消化液:a. 0.1%*:用0.1%CaCl 12(ph7.8)配制。b.0.4%*液:用0.1N的HCl配制。 6. 3%甲醇―H2O2溶液:用30%H2O2和80%甲醇溶液配制 7. 風裱劑:a. 甘油和0.5mmol/L碳酸鹽緩沖液(ph9.0–9.5)等量混合 b 油和TBS(PBS)配制 8.TBS/PBS PH9.0–9.5,適用于熒光纖維鏡標本;ph7.0-7.4適合光學纖維標本。 (三) 操作流程 1 脫蠟和水化:脫蠟前應將組織芯片在室溫中放置60分鐘或60℃恒溫箱中烘烤20分鐘。 a 組織芯片置于二甲苯中浸泡10分鐘,更換二甲苯后在浸泡10分鐘 b 無水乙醇中浸泡五分鐘 c 95%乙醇中浸泡五分鐘 d 75%乙醇中浸泡五分鐘 2 抗原修復:用于福爾馬林固定的石蠟包埋組織芯片: A 抗原熱修復 a 高壓熱修復 在沸水中加入EDTA(ph8.0)或0.01m*緩沖溶液(ph6.0).蓋上不銹鋼鍋蓋,但不能鎖定。將玻片置于金屬染色加上,緩慢加壓,是玻片在緩沖液中浸泡五分鐘,然后將蓋子鎖定,小閥門將會升起來。10分鐘后除去熱源,置入涼水中,當小閥門沉下去后打開蓋子。此方法適用于較難檢測或核抗原的抗原修復。 b 沸熱修復 電爐或水浴鍋加熱0.01*緩沖液(ph6.0)至95℃左右,放入組織芯片加熱10-15分鐘。 C 微波爐加熱 在微波爐里加熱0.01*緩沖液(ph6.0)至沸騰后將組織芯片放入,斷電,間隔5-10分鐘,反復1-2次。適用的抗原: Bcl-2.Bax.AR.PR.C-fos.x-jum.z-kit.c-myc.E-cadherin. ChromograninA.Cyclin.ER.Heatshock.Protein.HPV.Ki-67.MDMZ.P53.P34.P15.P-glycoprotein.PKC.PCNA.ras.Rb.等 B 酶消化方法 常用0.1%*和0.4%*液。*使用前預熱37℃,消化時間約為5-30分鐘。適用于被固定遮蔽的抗原:包括 Collagen.GFAP. Complement.Cytokeratin.C-erB-2.LCA.LN.等
