HLA Class II DR4抗體保存條件:Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

一、抗原抗體反應(yīng)特點(diǎn)
1.高度特異性,特異性是抗原抗體反應(yīng)的zui主要特征,特定抗原決定簇只與特定抗體結(jié)合;
2.可逆性:抗原與抗體結(jié)合形成抗原抗體復(fù)合物的過程是一種動(dòng)態(tài)平衡, 其反應(yīng)式為:Ag+AbAg·Ab;
3.適當(dāng)濃度和比例;
4.敏感性:在測(cè)定血清中某一物質(zhì)的含量時(shí),化學(xué)比色法的敏感度為mg/ml水平,酶反應(yīng)測(cè)定法的敏感度約為5~10μg/ml,免疫測(cè)定中凝膠擴(kuò)散法的敏感度與酶反應(yīng)法相仿。
二、影響抗原抗體反應(yīng)的因素
1.電解質(zhì) :抗原抗體特異結(jié)合后,膠體粒便可發(fā)生脫水作用,此時(shí)易受電解質(zhì)影響,如有適當(dāng)濃度的電解質(zhì)存在就會(huì)使它們失去部分負(fù)電荷而相互凝聚,于是出現(xiàn)明顯的凝集或沉淀現(xiàn)象。
2.酸堿度:合適的pH為6-8。
3.溫度:37℃通常是抗原體反應(yīng)zui適溫度。

免疫染色:
下面以石蠟切片的免疫組化為例演示詳細(xì)操作流程(SP法)
1.60℃烤片2小時(shí)
2.脫蠟。二甲苯脫蠟15分鐘,三次
3.水化。依次經(jīng)過無水乙醇、95%、90%、80%、70%,各5分鐘,蒸餾水(或自來水)5分鐘
4.PBS(洗液C-0079)洗5分鐘,三次
5.抗原修復(fù)。枸櫞酸緩沖液(修復(fù)液,C-0070)(約92-95℃)煮沸(沸水浴)修復(fù)15分鐘,自然涼至室溫。PBS洗5分鐘,三次
6.3%雙氧水(包含在SP-0023中)消化內(nèi)源性的過氧化物酶,20分鐘,PBS洗5分鐘,三次
7.正常山羊血清(包含在SP-0023中)封閉,37℃20分鐘
8.一抗孵育,4℃過夜. (抗體稀釋液C-0007)
9.PBS洗5分鐘,三次
10.二抗孵育,37℃20分鐘(二抗為*標(biāo)記的羊抗兔IgG,包含在SP-0023中),PBS洗5分鐘,三次
11.三抗孵育,37℃20分鐘(三抗為辣根酶標(biāo)記的鏈霉親和素,包含在SP-0023中),PBS洗5分鐘,三次
12.DAB(顯色劑C-0010)顯色,鏡下觀察結(jié)果,適時(shí)終止
13.蘇木素(C-0021)復(fù)染5分鐘,自來水沖洗片刻,1%的鹽酸酒精(75%)溶液(此試劑無法銷售,請(qǐng)自行配置,配制比例為:100ml 75%酒精+1ml濃鹽酸)分化30秒,自來水沖洗5分鐘藍(lán)化
14.脫水。依次經(jīng)過70%、80%、90%、95%、無水乙醇,各5分鐘,二甲苯15分鐘,三次
15.封片,中性樹膠(C-0073)封片

背景介紹:HLA-DRB1 belongs to the HLA class II beta chain paralogs. The class II molecule is a heterodimer consisting of an alpha (DRA) and a beta chain (DRB), both anchored in the membrane. It plays a central role in the immune system by presenting peptides derived from extracellular proteins. Class II molecules are expressed in antigen presenting cells (APC: B lymphocytes, dendritic cells, macrophages). The beta chain is approximay 26-28 kDa. It is encoded by 6 exons. Exon one encodes the leader peptide; exons 2 and 3 encode the two extracellular domains; exon 4 encodes the transmembrane domain; and exon 5 encodes the cytoplasmic tail. Within the DR molecule the beta chain contains all the polymorphisms specifying the peptide binding specificities. Hundreds of DRB1 alleles have been described and typing for these polymorphisms is routinely done for bone marrow and kidney transplantation. DRB1 is expressed at a level five times higher than its paralogs DRB3, DRB4 and DRB5. DRB1 is present in all individuals. Allelic variants of DRB1 are linked with either none or one of the genes DRB3, DRB4 and DRB5. There are 4 related pseudogenes: DRB2, DRB6, DRB7, DRB8 and DRB9.
