KLRA2蛋白抗體保存條件：Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

Western Blotting 實(shí)驗(yàn)步驟概述：

1、 組織取材：組織塊稱重

2、 利用液氮、研缽粉碎組織塊

3、 加入RIPA緩沖液（每克組織3ml RIPA），PMSF（每克組織30μl，10 mg/ml PMSF），

利用Polytron進(jìn)一步勻漿（15,000轉(zhuǎn)/分*1分鐘）維持4℃

4、 加入PMSF（每克組織30μl，10 mg/ml PMSF），冰上孵育30分鐘

5、 移入離心管4℃，約20,000g（約15,000轉(zhuǎn)）15分鐘

6、 上清液為細(xì)胞裂解液可分裝-20℃保存

7、 進(jìn)行Bradford比色法測(cè)定蛋白質(zhì)濃度

8、 取相同質(zhì)量的細(xì)胞裂解液（體積*蛋白質(zhì)濃度），并加等體積的2×電泳加樣緩沖液

9、 沸水浴中3分鐘

10、上樣

11、電泳（濃縮膠20mA，分離膠35mA）

12、電轉(zhuǎn)膜儀轉(zhuǎn)膜（100mA 40分鐘）

13、膜用麗春紅染色，膠用考馬斯亮藍(lán)染色

14、Westernblot 試劑盒顯色

背景介紹：The gene is a member of the large lectin-like type 2 transmembrane receptor family of the natural killer gene complex. The gene is located distantly omeric to its family's gene cluster on chromosome 6. The gene differs from the other genes in its cluster as its promoter region contains long and short interspersed repetitive elements suggesting a possible rearrangement or gene conversion. It is unknown whether this gene's encoded protein is involved with natural killer cell differentiation as are its other family members. Alternative splicing results in multiple transcript variants.
