跨膜受體蛋白Notch-3抗體保存條件：Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

跨膜受體蛋白Notch-3抗體,Anti-Notch3抗體免疫應答過程：

1.抗原進入機體；

2.抗原刺激 B淋巴細胞，使之分化增殖成為抗體分泌漿細胞和記憶細胞（初次免疫應答）；

3.抗體主要由漿細胞分泌 ；

4.記憶細胞再次受到特異性抗原刺激后會被激活而成為分泌抗體的漿細胞（再次免疫應答）；

5.記憶B細胞是長命細胞，可以存活數(shù)月甚至機體終生 。

Western Blotting 實驗步驟概述

1、 組織取材：組織塊稱重

2、 利用液氮、研缽粉碎組織塊

3、 加入RIPA緩沖液（每克組織3ml RIPA），PMSF（每克組織30μl，10 mg/ml PMSF），

利用Polytron進一步勻漿（15,000轉(zhuǎn)/分*1分鐘）維持4℃

4、 加入PMSF（每克組織30μl，10 mg/ml PMSF），冰上孵育30分鐘

5、 移入離心管4℃，約20,000g（約15,000轉(zhuǎn)）15分鐘

6、 上清液為細胞裂解液可分裝-20℃保存

7、 進行Bradford比色法測定蛋白質(zhì)濃度

8、 取相同質(zhì)量的細胞裂解液（體積*蛋白質(zhì)濃度），并加等體積的2×電泳加樣緩沖液

9、 沸水浴中3分鐘

10、上樣

11、電泳（濃縮膠20mA，分離膠35mA）

12、電轉(zhuǎn)膜儀轉(zhuǎn)膜（100mA 40分鐘）

13、膜用麗春紅染色，膠用考馬斯亮藍染色

14、Westernblot 試劑盒顯色

背景介紹：This gene encodes the third discovered human homologue of the Drosophilia melanogaster type I membrane protein notch. In Drosophilia, notch interaction with its cell-bound ligands (delta, serrate) establishes an intercellular signalling pathway that plays a key role in neural development. Homologues of the notch-ligands have also been identified in human, but precise interactions between these ligands and the human notch homologues remains to be determined. Mutations in NOTCH3 have been identified as the underlying cause of cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy (CADASIL).
