大鼠可溶性Toll樣受體2(sTLR2)免疫組化檢測試劑盒檢測方法組成結(jié)構(gòu)：
1、血清：操作過程中避免任何細(xì)胞刺激。使用不含熱原和內(nèi)毒素的試管。收集血液后，1000×g離心10分鐘將血紅細(xì)胞迅速小心地分離。
2、血漿：EDTA、檸檬酸鹽、肝素血漿可用于檢測。1000×g離心30分鐘去除顆粒。
3、細(xì)胞上清液：1000×g離心10分鐘去除顆粒和聚合物。
4、組織勻漿：將組織加入適量生理鹽水搗碎。1000×g離心10分鐘，取上清液。
5、豬基質(zhì)金屬蛋白酶9(MMP-9)酶免檢測盒供應(yīng)保存：如果樣品不立即使用，應(yīng)將其分成小部分-70℃保存，避免反復(fù)冷凍。盡可能的不要使用溶血或高血脂血。如果血清中大量顆粒，檢測前先離心或過濾。不要在37℃或更高的溫度加熱解凍。應(yīng)在室溫下解凍并確保樣品均勻地充分解凍。
注意事項(xiàng)
1. 當(dāng)混合蛋白溶液時(shí)應(yīng)盡量輕緩，避免起泡。
2. 洗滌過程非常重要，不充分的洗滌易造成假陽性。
3. 一次加樣時(shí)間控制在5分鐘內(nèi)，如標(biāo)本數(shù)量多，*使用排槍加樣。
4. 請每次測定的同時(shí)做標(biāo)準(zhǔn)曲線，做復(fù)孔。
5. 如標(biāo)本中待測物質(zhì)含量過高，請先稀釋后再測定，計(jì)算時(shí)請zui后乘以稀釋倍數(shù)。
6. 在配制標(biāo)準(zhǔn)品、檢測溶液工作液時(shí)，請以相應(yīng)的稀釋液配制，不能混淆。
7. 底物請避光保存。
8. 不要用其它生產(chǎn)廠家的試劑替換試劑盒中的試劑。
操作步驟：
1.編號：將樣品對應(yīng)微孔按序編號，每板應(yīng)設(shè)陰性對照 2 孔、陽性對照 2 孔、空白對照 1孔（空白對照孔不加樣品及酶標(biāo)試劑，其余各步操作相同）
2.加樣：分別在陰、陽性對照孔中加入陰性對照、陽性對照 50µl。然后在待測樣品孔先 加樣品稀釋液 40µl，然后再加待測樣品 10µl。加樣將樣品加于酶標(biāo)板孔底部，盡量不觸及孔壁，輕輕晃動(dòng)混勻，
3.溫育：用封板膜封板后置 37℃溫育 30 分鐘。
4.配液：將 30 倍濃縮洗滌液用蒸餾水 30 倍稀釋后備用
5.洗滌：小心揭掉封板膜，棄去液體，甩干，每孔加滿洗滌液，靜置 30 秒后棄去，如此 重復(fù) 5 次，拍干。
6.加酶：每孔加入酶標(biāo)試劑 50µl，空白孔除外。
7.溫育：操作同 3。
8.洗滌：操作同 5。
9.顯色：每孔先加入顯色劑 A50µl，再加入顯色劑 B50µl，輕輕震蕩混勻，37℃避光顯色
15 分鐘.
10.終止：每孔加終止液 50µl，終止反應(yīng)（此時(shí)藍(lán)色立轉(zhuǎn)黃色）。
11.測定：以空白空調(diào)零，450nm 波長依序測量各孔的吸光度（OD 值）。 測定應(yīng)在加終止 液后 15 分鐘以內(nèi)進(jìn)行。操作步驟：
