產(chǎn)品介紹：Steroid 5a-Reductase is an important enzyme in androgen physiology because it catalyzes the conversion of testosterone into the more potent 5a-dihydro-testosterone, which mediates androgen effects on target tissues. The enzyme exists as two isoforms: type 1, which is expressed mainly in the skin; and type 2, which is expressed mainly in the prostate. In cultured human skin cells, 5a-Reductase 1 shows heterogeneity of protein, and has different levels of transcriptional and translational expression. 5a-Reductase 1 is expressed in all portions of the hair follicle, whereas 5a-Reductase 2 is expressed only in mesenchymal portions. In addition, 5a-Reductase 1 is mainly expressed in human breast carcinoma and may play a role in the in situ production and actions of the potent androgen 5a-dihydrotestosterone, including inhibition of cancer cell proliferation in hormone-dependent human breast carcinoma. The 5a-Reductase-3a-hydroxysteroid dehydrogenase complex is present in the human brain, suggesting that the complex may be involved in the synthesis of neuroactive steroids or the catabolism of neurotoxic steroids.

Anti-SRD5A1抗體,類固醇5α還原酶1抗體進行免疫組化步驟：

石蠟包埋組織切片3~4μm 厚度
1.烤片：將待做切片置于切片架上，于60℃恒溫烤箱中至少烤1 hr；
2.脫蠟：切片放入盛有二甲苯的容器中脫蠟3次（即二甲苯Ⅰ、Ⅱ、Ⅲ），每次10 min；
3.水化：切片經(jīng)下行酒精水化，無水乙醇5min，95%乙醇2次（每次2min），85%乙醇2 min；75%乙醇2min，自來水沖洗，ddH2O洗2×2min；
4.抗原修復(fù)：根據(jù)抗體說明書*方法進行抗原修復(fù)，常采用高壓、微波（溫度達到98~100℃）或酶消化修復(fù)法，室溫自然冷卻，自來水沖洗，ddH2O洗2×2min，TBS洗滌（2×2min）（具體修復(fù)方法見附1）* 注：有些抗原勿需修復(fù)，直接進入第5步封閉。
5.封閉：滴加試劑B，37℃濕盒孵育30 min；
6.加一抗：滴加用試劑C（選擇合理的工作比例），37℃濕盒孵育2 hr 或4℃過夜；
7.洗滌： TBS-T洗滌（3×5 min）；
8.封閉：滴加試劑B，37℃濕盒孵育10 min；
9.加二抗：滴加用抗體稀釋液稀釋的*化二抗（試劑D），37℃濕盒中孵育30 min；
10.洗滌： TBS-T洗滌（3×5 min）；
11.封閉：滴加試劑Tween 20，37℃濕盒孵育封閉20 min；
12.加HRP-SA：滴加用試劑C稀釋的試劑E（1：50~200，終濃度5~20 nM），37℃濕盒中孵育30 min；
13.洗滌： TBS-T洗滌（3×5 min），TBS洗滌（2×5 min）；
14.顯色：應(yīng)用DAB溶液（試劑F）顯色；
15.復(fù)染：自來水充分沖洗，復(fù)染，脫水，透明；
16.封片：待組織標(biāo)本干后，用試劑緩沖甘油封固劑封片；
17.觀察成像：顯微鏡下觀察成像。
