 北京方程生物酶聯(lián)免疫吸附測(cè)定（enzyme-linked immunosorbent assay 簡(jiǎn)稱ELISA）是在免疫酶技術(shù)（immunoenzymatic techniques）的基礎(chǔ)上發(fā)展起來(lái)的一種新型的免疫測(cè)定技術(shù),ELISA過(guò)程包括抗原（抗體）吸附在固相載體上稱為包被，加待測(cè)抗體（抗原）, 再加相應(yīng)酶標(biāo)記抗體（抗原）,生成抗原（抗體）--待測(cè)抗體（抗原）--酶標(biāo)記抗體的復(fù)合物，再與該酶的底物反應(yīng)生成有色產(chǎn)物。借助分光光度計(jì)的光吸收計(jì)算抗體（抗原）的量。待測(cè)抗體（抗原）的定量與有色產(chǎn)生成正比。

elisa 小知識(shí)：
辣根過(guò)氧化物酶(HRP)是一種糖蛋白，每個(gè)分子含有一個(gè)氯化血紅素(protonhemin)區(qū)作輔基。酶的濃度和純度常以輔基的含量表示。氯化血紅素輔基的zui大吸收峰是403nm，HRP酶蛋白的zui大吸收峰是275nm，所以酶的濃度和純度計(jì)算式是(已知HRP的A(1cm 403nm 1%)＝25，式中1％指HRP百分濃度為100ml含酶蛋白1g，即10mg/ml，所以，酶濃度以 mg/ml 計(jì)算是HRP的A(1cm 403nm mg/ml=2.5)HRP純度(RZ)=A403nm/A275nm純度RZ(Ｒeinheit Zahl)值越大說(shuō)明酶內(nèi)所含雜質(zhì)越少。高純度HRP的RZ值在3.0左右，zui高可達(dá)3.4。用于ELISA檢測(cè)的HRP的RZ值要求在3.0以上。
