使用權(quán)限  A類培養(yǎng)需要滿足以下條件
1、無(wú)菌、無(wú)毒的環(huán)境：首先應(yīng)保證被培養(yǎng)的細(xì)胞處于無(wú)菌、無(wú)毒的環(huán)境，即對(duì)培養(yǎng)液和所有的培養(yǎng)用具進(jìn)行無(wú)菌處理。通常還要在培養(yǎng)液中添加一定量的抗生素以防培養(yǎng)過(guò)程中的污染。

2、營(yíng)養(yǎng)物質(zhì)：細(xì)胞體外培養(yǎng)所需營(yíng)養(yǎng)物質(zhì)與體內(nèi)基本相同，需要有葡萄糖、氨基酸、促生長(zhǎng)因子、無(wú)機(jī)鹽、微量元素等。通常還需加入血清、血漿等一些天然成分。

3、溫度和pH：細(xì)胞體外培養(yǎng)的適宜溫度一般與動(dòng)物的體溫相近。多數(shù)細(xì)胞生存的適宜pH為7.2～7.4。

4、氣體環(huán)境：培養(yǎng)所需氣體主要有O2和CO2，O2是細(xì)胞代謝所必需的，CO2的主要作用是維持培養(yǎng)液的pH。進(jìn)行細(xì)胞培養(yǎng)時(shí)，通常采用培養(yǎng)皿或松蓋培養(yǎng)瓶，將其置于含95%空氣加5% CO2的混合氣體的培養(yǎng)箱中進(jìn)行培養(yǎng)。

操作步驟：
1. 將無(wú)菌的蓋玻片置于細(xì)胞培養(yǎng)皿中，將細(xì)胞懸液接種于培養(yǎng)皿中進(jìn)行細(xì)胞爬片，待細(xì)胞長(zhǎng)至80%時(shí)取出爬片。
2. 用PBS浸洗爬片3次，4%的多聚甲醛固定爬片15min，再次用PBS浸洗玻片3次，每次3min。
3. 0.5% Triton X-100（PBS配制）室溫孵育爬片20min，使細(xì)胞通透。
4. 3% H2O2滴加爬片上以阻斷內(nèi)源性過(guò)氧化物酶，室溫孵育15min（一般用甲醇或蒸餾水稀釋30%過(guò)氧化氫），PBS沖洗3次，每次3min。
5. 封閉血清室溫孵育20min，PBS沖洗3次，每次3min。
6. 細(xì)胞特異性一抗孵育：按照抗體說(shuō)明書(shū)稀釋比例稀釋好一抗，滴加在爬片上，于4°C濕盒中孵育一夜，PBS沖洗3次，每次3min。
7. 二抗孵育：選與一抗對(duì)應(yīng)的二抗，按比例稀釋后滴加在爬片上，37°C濕盒中孵育30min，PBS沖洗3次，每次3min。
8. 將爬片周圍水漬吸干，爬片上滴加新鮮配制的DAB顯色液，顯微鏡下觀察，當(dāng)出現(xiàn)棕黃色或棕褐色的陽(yáng)性信號(hào)時(shí)用蒸餾水沖洗終止顯色。
9. 復(fù)染：用Harris蘇木素復(fù)染30s~1min，水洗后用1%的鹽酸酒精分化，再用蒸餾水（或PBS）水洗返藍(lán)。
10. 封片：將中性樹(shù)膠滴在爬片一側(cè)，再用蓋玻片蓋上，先放平蓋玻片一側(cè)再輕輕放下另一側(cè)，以免產(chǎn)生氣泡，封好的爬片平放置于通風(fēng)廚中晾干。
11. 鏡檢觀察。

Concanaalin A (Con A) Agarose consists of Con A coupled to 4% agarose by the cyanogen bromide method. Con A is a tetrameric metalloprotein lectin isolated from Canaalia ensiformis (jack bean). Con A is used for the purification of glycoproteins, polysaccharides and glycolipids as it binds molecules containing α- D-mannopyranosyl, α-D-glucopyranosyl and sterically related residues. Con A agarose has also be used in other application areas including purification of enzyme-antibody conjugates, purification of IgM and separation of membrane esicles.
